RESUMO

We provide a morphological and molecular-based description of the eumonostiliferous hoplonemertean Oerstedia fuscosparsa sp. nov. based on two specimens dredged from a depth between 11 and 18 m off Kouyatsu, Tateyama, Chiba, Japan. This Oerstedia differs from its congeners by having a deep brownish pigmentation in the head, four orange ocelli, and imprecise dorsal and ventral transverse bands composed of numerous small brown spots. One of the specimens, herein designated as the holotype, has deep brown spots situated mid-dorsally onto the transverse bands running throughout the body. These dorsal markings are absent in the other specimen designated as a paratype. Internally, the species has accessory lateral nerves, and the proboscis nerves are not distinct. Molecular phylogenetic analyses based on partial sequences of the 16S, 18S, 28S rRNA, cytochrome c oxidase subunit I, and histone H3 gene markers along with the sequences from 22 Oerstedia species available in public databases confirmed that our species is sister to Oerstedia phoresiae (Kulikova, 1987) and belongs to the recently established Paroerstediella clade within the genus Oerstedia.

RESUMO

A new species of amphinomid polychaete, Branchamphinome kohtsukai sp. nov., is described from Japanese waters, 29-211 m in depth. The species is distinguishable from other congeners by the following features: i) branchiae with four-six filaments in midbody segments; ii) two pairs of eyes not coalescent; iii) the ventral side of the first four chaetigers broadly pigmented. This is the first record of Branchamphinome from the North Pacific Ocean. We provide a phylogenetic tree based on the sequences of four genes (COI, 16S, 18S, 28S) and discuss amphinomids' relationships.

RESUMO

Morphological and molecular analyses of Korean rhizocephalan barnacle species were performed to examine their host ranges and taxonomy. Morphological examination and molecular analysis of mtDNA cox1, 16S, and nuclear 18S rRNA sequences revealed nine rhizocephalan species from three genera of the two families, Sacculinidae and Polyascidae. Phylogenetic analysis of molecular sequences revealed two new species candidates in the genus Parasacculina, and three Sacculina species (S. pilosella, S. pinnotherae, and S. imberbis) were transferred to the genus Parasacculina. Examination of host ranges revealed higher host specificity and lower infestation rates in Korean rhizocephalan species than rhizocephalans from other geographic regions. This is the first report of the taxonomy, species diversity, and host ranges of Korean parasitic rhizocephalan barnacles based on their morphological and molecular analyses. More information from extensive sampling of parasitic barnacles from a wide range of crustacean host species is necessary to fully understand their taxonomy, prevalence on decapod hosts, and phylogenetic relationships among major rhizocephalan taxa.

RESUMO

A new species of diogenid hermit crab, Diogenes minimus, is described and illustrated on the basis of material from shallow subtidal waters (5-19 m) in central Japan. It appears close to D. holthuisi Asakura Tachikawa, 2010, but easily distinguished from the latter by the shape of the antennal acicle, non-elongate male left cheliped and the armature of the right cheliped merus. Including the new species, 14 identified species of Diogenes are now known from Japanese waters. A brief overview on species of Diogenes recorded from Japanese waters is given.

We performed a diversity study on parasitic barnacles (Crustacea: Cirripedia: Rhizocephala: Peltogastridae) that parasitize hermit crabs in Korea. Their morphological, ecological, molecular (cytochrome c oxidase subunit I, 16S rRNA), and biogeographical characteristics were examined. Three species were identified based on GenBank sequences and the external morphology of the externa. In addition, this study proposes four new candidate species. This is the first report on the family Peltogastridae from Korea. Six hermit crab species were found to be new hosts to peltogastrids. Korean peltogastrids are less prevalent on their host hermit crabs than those from Japan are, especially in the west coast of Korea. Peltogasterella gracilis is widely distributed throughout Korea, Peltogaster lineata is located on the east coast, and Peltogaster postica is only located on Jeju Island.

Coral bleaching can be induced by many different stressors, however, the most common cause of mass bleaching in the field is higher than average sea surface temperatures (SST). Here, we describe an unusual bleaching event that followed very calm sea conditions combined with higher than average SST. Patterns of mortality differed from typical bleaching in four ways: 1) mortality was very rapid; 2) a different suite of species were most affected; 3) tissue mortality in Acropora spp. was often restricted to the center of the colony; 4) the event occurred early in summer. The two weeks prior to the event included 8 days where the average wind speed was less than 3 ms -1. In addition, SSTs in the weeks preceding and during the event were 1.0-1.5°C higher than the mean for the last 30 years. We hypothesize that this unusual bleaching event was caused by anoxia resulting from a lack of water movement induced by low wind speeds combined with high SST.

A new rhizocephalan species, Peltogaster unigibba n. sp., is described from the host hermit crab, Pagurixus boninensis (Melin, 1939), from the Bonin Islands, Japan. Of the16 known species of Peltogaster now currently recognised, P. unigibba n. sp., and P. contorta Boschma, 1958 share a left lobe that projects beyond the mantle aperture. The two species can be distinguished from one another by the position of the opening of the mantle aperture. The new species most closely resembles P. naushonensis Reinhard, 1946 in its internal structure, but clearly differs in the relative length of the colleteric glands. Peltogaster unigibba n. sp. represents the first record of a rhizocephalan from the oceanic Bonin Islands, and the second record of a rhizocephalan from an oceanic island in the northern hemisphere.

Despite the presence of ß-1,2-glucan in nature, few ß-1,2-glucan degrading enzymes have been reported to date. Recently, the Lin1839 protein from Listeria innocua was identified as a 1,2-ß-oligoglucan phosphorylase. Since the adjacent lin1840 gene in the gene cluster encodes a putative glycoside hydrolase family 3 ß-glucosidase, we hypothesized that Lin1840 is also involved in ß-1,2-glucan dissimilation. Here we report the functional and structural analysis of Lin1840. A recombinant Lin1840 protein (Lin1840r) showed the highest hydrolytic activity toward sophorose (Glc-ß-1,2-Glc) among ß-1,2-glucooligosaccharides, suggesting that Lin1840 is a ß-glucosidase involved in sophorose degradation. The enzyme also rapidly hydrolyzed laminaribiose (ß-1,3), but not cellobiose (ß-1,4) or gentiobiose (ß-1,6) among ß-linked gluco-disaccharides. We determined the crystal structures of Lin1840r in complexes with sophorose and laminaribiose as productive binding forms. In these structures, Arg572 forms many hydrogen bonds with sophorose and laminaribiose at subsite +1, which seems to be a key factor for substrate selectivity. The opposite side of subsite +1 from Arg572 is connected to a large empty space appearing to be subsite +2 for the binding of sophorotriose (Glc-ß-1,2-Glc-ß-1,2-Glc) in spite of the higher Km value for sophorotriose than that for sophorose. The conformations of sophorose and laminaribiose are almost the same on the Arg572 side but differ on the subsite +2 side that provides no interaction with a substrate. Therefore, Lin1840r is unable to distinguish between sophorose and laminaribiose as substrates. These results provide the first mechanistic insights into ß-1,2-glucooligosaccharide recognition by ß-glucosidase.

Peltogasterella sensuru n. sp. infests Pagurixus hermit crabs inhabiting rocky shores off Okinawa Island (Ryukyu Archipelago, Japan). This species is clearly distinguishable from Peltogasterella gracilis (Boschma, 1927): the stalk emerges from the middle part of the externa in the present species and P. sulcata (Lilljeborg, 1859), while the stalk in P. gracilis emerges from the posterior end of the externae. The new species differs from P. sulcata based on the morphology of the mantle aperture. Peltogasterella sensuru n. sp. repeatedly produces single brood externae that have not been previously observed in species belonging to the suborder Kentrogonida Delage, 1884. We also determined partial sequences of the COI gene and 16S rRNA gene of the new species for use as molecular markers for species identification.

Lin1840 is a putative ß-glucosidase that is predicted to be involved in 1,2-ß-glucan metabolism since the lin1839 gene encoding a 1,2-ß-oligoglucan phosphorylase and the lin1840 gene are located in the same gene cluster. Here, Lin1840 was crystallized. The crystals of Lin1840 diffracted to beyond 1.8 Šresolution. The crystal belonged to space group I121, with unit-cell parameters a = 89.75, b = 95.10, c = 215.00 Å, α = 90.00, ß = 96.34, γ = 90.00°.

A new genus and two new species of Peltogastridae, Peltogaster postica sp. nov. and Ommatogaster nana gen. et sp. nov., are described from Okinawa Island, Ryukyu Islands, southwestern Japan. The two new rhizocephalans were found to be parasitic on the estuarine hermit crabs, Pagurus minutus Hess, 1865 and Diogenes leptocerus Forest, 1956, respectively. Peltogaster postica sp. nov. is allied to P. curvata Kossmann, 1874, P. paguri Rathke, 1842 , and P. reticulata Shiino, 1943 , but is distinguished by its relative length and internal and external structures of the mature externa. Ommatogaster gen. nov. is established for the present new species O. nana based on the morphologies of the visceral mass of the externa and the presence of a nauplius eye in the larvae. Partial COI sequences were obtained from the two new species and one known species, Dipterosaccus indicus Van Kampen and Boschma, 1925, to test the possible usefulness of the sequences as tags for species identification.
